Dorothy Gillespie was born in Roanoke VA. She studied at the Maryland Institute College of Art in Baltimore, MD and at the Art Student's League in New York, NY.

Gillespie was an Artist in Residence, Women's Interart Center in New York, NY in 1972 and had a lecture series at the New School for Social Research, New York, NY in 1977. She was a Professor of Art at Radford University, Radford, VA from 1997-99; on the Board of Trustees, Maryland Institute, College of Art in Baltimore, MD from 1996-99; on the Board of Trustees of the Maitland Art Center, Maitland, FL from 1996-99. She was also a Woodrow Wilson Visiting Fellow from 1985-1993, where she visited small private colleges throughout the U.S., gave lectures and coached and taught student artists. Gillespie published a book through the Radford University Foundation Press. She is best known for her large, colorful abstractions. In 2012, Gillespie died in Coral Gables, FL at the age of 92.
